- Introduction
While taking a brain science course during the summer camp at a university, I learned that there is a certain drug dose for a drug to efficiently reach its target place in our bodies. Curious of how the medicine or drug dose is determined, I learned that the partition coefficient of drugs plays an important role in determining drug dose. Contrary to my expectation, drugs are not distributed evenly throughout a body after intake. This is because our cell membranes, which are consisted of lipid bilayers, allow only some solutes to pass while blocking others. Lipid-soluble (nonpolar) molecules can permeate the membrane, but the lipid bilayer repels water-soluble (polar) molecules.[1] Thus, lipophilicity [2], which is measured by partition coefficient, is a major structural factor governing pharmacodynamics of drugs. Different partition coefficients lead to different distribution patterns; hydrophobic drugs with high partition coefficients are mainly distributed to hydrophobic areas such as blood brain barrier (“Blood-Brain Barrier”), and hydrophilic drugs with low partition coefficient are distributed to aqueous areas such as blood serum (“Blood-Brain Barrier”)…
